I currently have a board which is used for project tracking. The primary column is project number, which currently manually inputted. I want to make it so when a new project is added, it auto generates the project number following on from the previous number - is this possible?

Are you open to using a different column? There is a column that does that called ‘Auto Number’.

You can also use the app AutoID Column from the marketplace that can write a generated ID (that is configurable) to any text column or the leftmost (name) column.

Along with @Clover_Gal ‘s suggestion, you can also use the Item ID column and customize it to your liking. Mind you, you can also “kind of” do the same thing with the Auto number column.
The app does offer more functionality if needed. Otherwise I recommend the ItemID column. The issue with AutoNumber column is that the numbers gets reassigned when items are ordered differently. With AutoNumber the number does not stay with the item when the item is moved. Using the ItemID column the ID will stay with the item as long as the item is moved to somewhere in the same board. When you have the requirement that the ID always stays with the item the mentioned app is the way to go.
What I do is use Monday’s item ID and customize it and when it leaves to another board, I have it mapped in to come in as a text column Keeping the same project ID throughout the entire lifecycle of the project
