Automation to Notify Users Supervisor Listed under the Users Profile

We are requesting a New Automation to Notify an Item Users Supervisor if an Item is Overdue. We would like the Supervisor to be pulled from the Item Users Profiles. We do not want to add an additional column labeled “Supervisor” on each Board we create.
